The Worm Lysis Kit is designed for rapid, efficient, column-free extraction of PCR-ready DNA from C. elegans. Traditional extraction methods take 2 hours, the Worm Lysis Kit requires only a 15-minute incubation before the DNA is ready for use directly in your PCR.

DNA extraction is performed in a single tube without multiple washing steps to reduce potential contamination and sample loss and allows for quality DNA templates to be obtained consistently from single or multiple worm lysis reactions.

sqt-1 marker plasmid (pNU1228)

The sqt-1 plasmid encodes for a cuticle collagen expression in C elegans.

Composition is sqt-1p::SQT-1(genomic)::sqt-1utr.

A single point mutation (e1350) within the gene produces a morphological phenotype similar to Rol due to an improper cuticle formation.

Expression of SQT-1 is seen during L2, L3, and L4 larvae and adult stages, but not L1 or dauer larvae.

This plasmid is suggested as a co-marker of injection efficiency in C. elegans and other nematodes. Successful injections will display a Rol phenotype in L2-L4 larvae and adult worms. In the injection mix, a final concentration is commonly used at 10 ng/µL in a 100ng/µL total DNA. The reagent is supplied as a 10X stock.

 

myo-2::GFP marker plasmid (pNU675)

The myo-2::GFP construct expresses GFP in the pharynx in C. elegans.

Composition is myo-2p::eGFP(3x-syntrons)::tbb-2utr.

The myo-2 promoter drives GFP exclusively in pharynx of the animal.

This plasmid can be used as as a co-marker of injection efficiency in C. elegans or other similar nematodes. Successful injections will display a bright green pharynx. In the injection mix, a final concentration of 1.5 ng/µL is commonly used in a 100ng/µl total DNA. The reagent is supplied as a 10X stock. 

 

hygR selection plasmid (pNU1254)

The HygR plasmid encodes for hygromycin resistance, a useful selection tool, resulting in broad expression of hygromycin B phosphotransferase (HPT) in C. elegans tissues.

The HygR plasmid is commonly added into an injection mix at a final concentration of 10 ng/µL in a 100 ng/µL total DNA mix.

Sufficient accumulation of HPT expression occurs 24 hours post injection and survival starting two days post Hygromycin B exposure (0.25 mg/mL) indicates resistance.

By day five of post hygromycin exposure, only resistant animals are present.

This HygR plasmid is a good selection tool for use in C. elegans or other similar nematodes. Successful injections will be resistant to Hygromycin B (0.25mg/mL) in plates. Addition of HygR plasmid into an injection mix is commonly used at a final concentration of 10 ng/µL in a 100ng/µL total DNA. The reagent is supplied as a 10X stock.

Description

Product Information

A common way to test what proteins do in biological processes is to perform loss-of-function assays. Phenotypic readouts of loss-function cells/organisms give indications of what a specific protein function does.

Fusing GFP (or another fluorophore) to the gene of interest (GOI) enables visualization of the cells and sublocalization of the GOI. However, without modulation of protein expression one cannot know what the function of the protein is. Fluorescent protein RNAi bacteria can be used to knock down any protein that is fused to GFP. It’s designed to inducibly express interfering RNA (RNAi) that targets GFP mRNA for degradation. And It can be fed to C. elegans as their food source so they are exposed to the RNAi as they eat and grow.

  • Reduce expression of anything GFP tagged instead of individual RNAi bacterial strains for each gene
  • Use the exact same strain and genetic background as an experimental control for functional studies
  • Knockdown your endogenous protein of interest

In CRISP GFP insertion lines, progeny of L4/adult worms exposed to induced GFP RNAi bacteria lose expression of their gene of interest (GOI) that is fused to GFP. L4/Adult GFP inserted worms were plated on either control (HB101) or GFP RNAi bacteria. After 48hrs, eggs were imaged for GFP. GFP RNAi treated eggs do not express GFP, while control eggs do.

L4/Adult GFP-inserted worms were plated on either control (HB101) or GFP RNAi bacteria. After 4 days, an image of the bacteria seed was imaged. In this example, GFP is fused to hrp-1. RNAi against hrp-1 is lethal. This indicates that depleting a GFP fused form is also lethal.
